You have Windows 2000 installed on a USB Thumb/Flash drive and want to boot to it instead of the HDD/Hard Disk Drive you have Win8 installed on? It probably will work as a Win8 computer is usually new enough to boot to a USB device and it will be as safe as booting to the HDD as far as infections are concerned. At issue will be whether Win2000 can support the devices on the motherboard, have drivers for them such as Sound, Video, Network Adapter, etc. But do keep in mind there's been a lot of advances in technology since Win2000 faded away in favor of WinXP.

Something else to bear in mind - if you boot from a USB, your OS will run at USB speeds i.e.. very slowly, especially if its USB2.
